          <dc:title>Partial Purification of Cathepsin D from Muscle of Chum Salmon in Spawning Migration</dc:title>

          <dc:description>Acid proteases such as cathepsin D or similar protease(s) have previously been ascribed to be one of the causes of softening or lyzing phenomenon of the muscle which sometimes observed in chum salmon caught during spawnig migration. In this study, separation of acid protease(s) from skeletal muscle of both salmon caught during spawning migration and feeding migration was attempted. Only one fraction containing acid proteases was obtained from the respective specimens by following procedures: acid treatment of 0.2 M KCI crude muscle extract, warming the extract at neutral pH, DEAE-Sephadex chromatography and gel-filtration on Sephadex G 100 column. A 300 to 400-fold purification with about 50% recovery was attained. The properties of both perotease preparations substantially coincided with those of cathepsin D.</dc:description>
          <dc:description>産卵回帰中に漁獲されたシロザケ Oncorhynchus keta 筋肉から,それらにしばしばみられる筋肉の軟化·融解現象への関与が大きいと考えられたカテプシンDを分離した。その精製度合は,抽出液の比活性に対し322倍,収量は54%であった。索餌回遊中漁獲されたシロザケ筋肉からも同様にしてカテプシンDを,精製度合392倍,収量49%で分離した。両者からの画分中のカテプシンDの性質は調べた限りでは互いに一致した。すなわち,分子量は,4.1-4.2×10⁴ へモグロビン分解の至適 pH(2時間反応)は3.4付近であり,leupeptin では阻害されず,pepstatin で完全に阻害された。</dc:description>
