This EIA system used a horseradish peroxidase-insulin complex as a labeled hormone and 3-(p-hydroxyphenyl) propionic acid as a fluorescent substrate of peroxidase. The specificity of this EIA was proved by the facts that no cross-reactions were detected with mammalian pancreatic hormones except for insulin. The minimal detectable amount in this EIA system was 12 pg/well for bovine insulin, and the sensitivity was higher than those of the systems previously developed for measuring mammalian insulin. Walleye pollock (Theragra chalcogramma) insulins were detected in HPLC fractions by this EIA system. Although we purified walleye pollock insulins from only 230 mg Brockmann bodies, we could obtain enough to analyze the amino acid sequence. The results indicate that the sensitivity of this EIA system is sufficiently high for small-scale purification of fish insulin. However, the estimated amounts of walleye pollock insulins by this EIA system were considerably lower than the assumed amounts from their ultraviolet absorbance.
