内容記述 It is a well known phenomena in abalone hatchery that the egg membrane dissolved and the embryonic development became abnormal in dense sperm water and also that the fertilization rate was a low level in sparse sperm water. In the present paper, the reasonable sperm density for the artificial fertilization of abalone is dealt with. Adult abalones, Haliotis discus hannai INO, were reared in indoor tanks as reported previously (KIKUCHI and UKI, 1974a). In the experiment, they had an integral temperature of 1,043. One female and two males spawned almost at the same time artificially by the method of 2,537A U.V. irradiated sea water (KIKUCHI and UKI, 1974b). Sperm waters of several density levels were prepared soon after spawning as shown in Table 1, and each 50 ml of the sperm water was kept in glass tubes. Eggs, 1,000 to 5,000 in number, were then put in the glass tubes for fertilization. The fertilization rate was observed after four hours thereafter, and the embryonic development was observed constantly. The results are as follows. The fertilization rate showed a low level in sparse sperm water such as 1,000 sperm per ml. By increasing the sperm density, the rate advanced and reached almost 100 percent of the fertilization rate in 100,000 per ml. The egg membrane dissolved in 1,930,000 per ml and the embryo showed abnormal development in 4,830,000 per ml (Figs. 1 and 2). From these facts, it may be considered that the reasonable sperm density for fertilization of the abalone is from 100,000 to less than 1,900,000 per ml. As technique, it seems to be desirable that the sperm density is 200,000 per ml in order to obtain a suitable artificial fertilization and to decrease the surplus sperm which induced the abnormal phenomena in the early development of the abalone. During the experiment, an apparatus for trial was made as shown in Fig. 3 using photo-absorbancy. The accuracy of absorbancy was confirmed as shown in Fig. 4. It is useful for easy measuring of the sperm density.
