Ovarian pieces consisting of about 70 oocytes with follicular tissues (ca. 50 mg) in the pre-, middle-, and post-stages of germinal vesicle migration (GVM) respectively were incubated on a rotary shaker under 100% O2 atmosphere, with densities varying from 8 mg/ml to 675 mg/ml, in medium 199 (with EARLE's BSS) for 18 hours at 25℃. Spontaneous GV breakdown (GVBD, i.e. beginning of meiosis) increased to some extent with the advancement of GVM, and in proportion to the incubation density of the ovarian pieces up to 225 mg/ml. At 10 μg/ml, which was the most efficient dose, sc-EPa significantly induced GVBD with any combination of the GVM stage and the incubation density. However at 1μg/ml and 100μg/ml, sc-EPa failed to induce GVBD when the oocytes in the preand middle-GVM stages were used under the low inducbation density.
