| 内容記述 | Effects of solvent systems and activation of plate were studied on TLC separation of nalidixic acid (NA), oxolinic acid (OA), piromidic acid (PA), unconjugated (α-OHPA, β-OHPA) and conjugated metabolites of these drugs in rainbow trout. Drugs were introduced into rainbow trout orally. Biles were withdrawn 24 hr later, while urines were collected for 48 hr. After extraction of sample fluids with chloroform, unconjugated and conjugated substances were separated into chloroform and aqueous layer, respectively. TLC of unconjugated substances were examined using both standard and chloroform layer of extracts, and TLC of conjugated drugs using aqueous layer. Six acidic solvent systems including n-BuOH or CHCl₃ systems, a neutral solvent system and a basic solvent system were examined. Among the solvent systems examined n-BuOH-AcOH-H₂O (4: 1: 1) produced the best separation for NA and OA, and CHCI₃-formic acid-ethyl formate (8: 1: 4) for PA, α-OHPA and β-OHPA in chloroform layer, respectively. For the sepatation of conjugated NA, OA and PA in aqueous layer n-BuOH-AcOH-H₂O (4: 1: 1) was found to be the most efficient. Little difference of Rf value was observed between the separation of NA and OA using activated and unactivated plate. However, better sepatation was obtained for PA, α-OHPA and β-OHPA using unactivated plate in high humidity of developing chamber. | |||||
