| 内容記述 | The crude enzyme extract was found to be successfully recovered from the liver homogenate of Japanese common squid Todarodes pacificus using cooled acetone (-20℃) for the removal of liver lipid, and 0.05 M sodium acetate buffer (pH 4.0) for the protease extraction from its acetone powder. Among the Japanese common squid, flying squid Ommastrephes bartrami, arrow squid Loligo bleekeri, cuttlefish Sepia esculenta, and diamondback squid Thysanoteuthis rhombus, the crude enzyme extract from the diamondback squid showed the highest proteolytic activity at both pH 4.0 and 7.0. Especially, its activity at pH 7.0 was 2.4-7.6 times higher than those of the other species, although all of the enzyme extracts showed higher activity at pH 4.0. It is also suggested that the crude enzyme extract from diamond-back squid contains at least three kinds of proteases based on SP-Sephadex C-25 ion-exchange chromatography. | |||||
