Hybrids derived from a cross between carp and Carassius fish containing the genome of crucian carp and goldfish have been artificially produced for use as food and aquarium fish. We obtained a third generation of diploid and triploid hybrids(F3)derived from crosses between the F2 hybrid of family 1[F1 hybrid(C. carpio ♀ × Carassius cuvieri ♂)♀ × C. carpio ♂]♀ and C. auratus ♂ and cross between the F2 hybrid of family 2[F1 hybrid(C. carpio ♀ × C. cuvieri ♂)♀ × C. cuvieri ♂ ] ♀ and C. auratus ♂. However, it is difficult to estimate the genome composition of the F3 hybrids by morphological measurement. We previously developed three methods of species distinction between carp and Carassius fish:(1)analysis by PCR amplification using carp- and crucian carp-specific primers targeting the IGF-1 gene region,(2)PCR-RFLP analysis of the TGF-beta gene region, and(3)PCR-RFLP analysis of the mitochondrial 16S ribosomal RNA gene region. In this study, we estimated the genome composition of eight F3 hybrids by these methods. We estimated that five F3 hybrids contained the genomes of C. carpio, C. cuvieri, and C. auratus. Thus, the five F3 hybrids are considered to be allotriploids and include the genomes of three fish species. We estimated that two F3 hybrids contained the genomes of C. carpio and C. auratus. We also estimated that one F3 hybrid contained the genomes of C. cuvieri and C. auratus . Thus, these three F3 hybrid types are considered to be diploid hybrids.
