| 内容記述 | The purpose of this study is to develop a simple method for short term preservation of spermatozoa in the barfin flounder Verasper moseri and the spotted halibut Verasper variegatus . Firstable, artificial seminal plasma (ASP) was prepared based on some chemical and physical properties of the seminal plasma of both fish. Fertility and motility of both spermatozoa preserved as milt and spermatozoa preserved after dilution with the ASP were examined. In the barfin flounder, spermatozoa preserved as milt could maintain normal fertility and motility for 4 days. Spermatozoa preserved after dilution with ASP could also maintain normal fertility for 4 days, although, sperm motility was immediately reduced until first day of the preservation. Motility of spermatozoa preserved after dilution with the ASP was recovered in sea water which was incubated with egg of the barfin flounder (egg SW). This result suggests that the egg SW has sperm motility activating factor, and may explain why the spermatozoa could maintain their fertility. Fertility of spermatozoa preserved after diltuion with the ASP tended to be more stable than those preserved as milt. In the spotted halibut, spermatozoa preserved after dilution with the ASP could also maintain normal fertility for 3 days. These results suggest that it is practicable for short term to preserve spermatozoa with dilution in the ASP. | |||||||||||||||||||||||||||||||||||||||||
