| 内容記述 | This report deals with the accuracy of measuring K value, an index for estimating freshness of fish, by freshness testing paper (FTP) method, which was developed as a simple and rapid method for estimating freshness of fish. In the procedure of the FTP method, inosine and hypoxanthine are determined, but the total amount of ATP and its related compounds is not determined by assuming it to be definite in a given species of fish. Therefore the accuracy of the FTP method was compared with that of a usual enzymatic method and a simple column chromatography reported by UCHIYAMA et al. Carp and bigeye tuna were used as sample and K value of same extract was measured by different methods. Absorbances of the muscle extract of 17 fish species were measured and the difference of the content of ATP and its related compounds among fish in a species was estimated. The results obtained were as follows ; 1) A coefficient of correlation between the values obtained by the FTP method and those by the column chromatography was smaller than one between the values obtained by the enzymatic method and the column chromatography (Figs, 1 and 2). 2) K value measured by the FTP method was widely scattered, its confidence interval reaching ±ca 15% against its regression line to the values measured by the column chromatography, while K value measured by the enzymatic method scattered narrowly, its confidence interval was limited to ± ca 5% against its regression line (Figs. 1 and 2). 3) When mean K value measured by the column chromatography were 9.3% and 56. 2%, mean K value measured by the FTP method deviated 7.6% and 9.4% from them, respectively, while mean K value measured by the enzymatic method deviated 1.1% and 2.8%, respectively (Table 1). 4) The accuracy, viewed from the coefficient of variance, of the FTP method was 1/2-1/4 of the other 2 methods (Table 1). 5) The ratio of the difference between the maximum and the minimum values, in a species, of absorbance at 250 nm of muscle extract to the maximum value reached more than 30% in 11 species and more than 50% in 4 species out of 17 fish species investigated. The content of ATP and its related compounds in muscle varied from fish to fish even within a species (Fig. 4). 6) It was concluded that the deviation of the K value measured by the FTP method would be caused by not only the variation, in a species, of the content of ATP and its related compounds in muscle but also the method per se for determining inosine and hypoxanthine by using the colour chart for converting into K value (Figs. 4, 6, 7, and Table 2). | |||||||||||||||||

| 内容記述 | 生鮮度試験紙(FTP)によるK 値測定精度を改良簡易カラムクロマトグラフィーおよび酵素法の精度と比較した。また,筋肉の ATP関連化合物の個体差あるいは部位差を推定するため,17魚種について抽出液の 250nm 吸光度を測定した。1) FTPによる測定値は,カラム法の測定値との間に有意な相関(r=0.701 ** ,n=29)が認められたがが,酵素法の測定値とカラム法の測定値との相関係数(r=0.971 ** ,n=30)より小さかった(Figs.1,2)。2) FTPによる測定値はバラツキが大きく,その信頼区間は,カラム法の測定値との回帰直線に対し,±15%に達した。一方,酵素法ではパラッキが小さく,測定値の信頼区間は回帰直線に対し,±5%以内であった。3) カラム法の測定値平均が9.3%および56.2%の筋肉抽出液では,酵素法およびカラム法による測定値平均値の差はそれぞれ,1.1%および2.8%であったが,FTP法およびカラム法による測定値平均値の差はそれぞれ,7.6%および9.4%に達した。4)FTP法の,変動係数からみた,精度はカラム法および酵素法の1/2~1/4であった(Table1)。5) 筋肉抽出液の250nm吸光度の最大値と最小値の差が,最大値の30%を越える例が調査魚種17例中,11例見られ,うち4例は50%を越えた。すなわち,筋肉ATP関連化合物含量は同一魚種でも一定ではない。6) FTP法によるK値測定精度がカラム法および酵素法に比べて劣る主要な原因は,ATP関連化合物の個体差あるいは部位差を無視して,一魚種一定と定めて測定することの他に,K値換算色標によるイノシソ,ヒボキサンチンの測定それ自体にもあると考察された(Figs.4,6,7,Table 2)。 | |||||||||||||||||
