| 内容記述 | Excessively soft or lyzed meats are sometimes observed in chum salmon trapped by a set-net just befor entering the spawning streams, posing serious problems in the coastal salmon fisheries industry. This abnormality has previously been presumed to be caused by proteolysis of structural proteins in the muscle tissue, based on the fact that the level of protease activity in the muscle of chum salmon caught during spawning migration is always several times higher than that of chum salmon caught during feeding migration. The present study was undertaken to elucidate which type of protease is attributable to the muscle protein degradation. Proteases were differentiated by inhibition experiments with leupeptin and pepstatin. Autolytic activity of both muscle homogenate and muscle extract prepared from the salmon caught during spawning migration were strongly inhibited by leupeptin and weakly by pepstatin, and inhibition of autolytic activity of the well-washed myofibrillar fragments was the reverse case. Hemoglobin hydrolyzing activity at pH 3 and casein-hydrolyzing activity at pH 6 of the muscle extract were completely inhibited by pepstatin but not at all by leupeptin. These findings imply that proteolysis of myofibrils, in other words, muscle tissue degradation was mainly brought about by acid protease(s) such as cathepsin D. Inhibition experiments also indicated the presence of at least two other kinds of proteases, one of them was a thiol protease which hydrolyzed chiefly sarcoplasmic proteins in the low pH region, and the other was an acid protease which hydrolyzed casein used as substrate in the slightly acidic pH region, possibly differing from cathepsin D. | |||||

| 内容記述 | 産卵のため回帰するシロザケにしばしばみられる筋肉(普通肉)の軟化·融解現象に,同筋肉に活性の高いカテプシンDあるいはD様プロテアーゼがかなり深く関与している,すなわち,筋原繊維を分解するものと推定された。他に,強い酸性下で筋形質タンパク質をよく分解する一種のチオールプロテアーゼ,および弱酸性下で基質カゼインをよく分解する一種の酸性プロテアーゼの存在も示唆された。回帰ザケ筋肉の高プロテアーゼ活性は,活性に影響する factor によるものでなく,プロテアーゼの濃度が高いことによるものと考えられた。 | |||||
