| 内容記述 | In order to develop a method for long-term storage, protoplasts isolated from thalli of Porphyra yezoensis form. narawaensis were frozen using a two step procedure with an initial slow cooling rate (1℃/min) to - 40℃ followed by a rapid cooling rate when samples were plunged into liquid nitrogen. The addition of 0.7 M mannitol as a cryoprotectant and prefreezing treatment were necessary for successful cryopreservation. About 70% of the protoplasts were alive after the freeze-thaw cycle using this method. | |||||
