| 内容記述 | May-Grünwald Giemsa and Giemsa staining properties of cytoplasmic granules in carp neutrophils were reexamined in this study. Three types of granules; eosinophilic granule (αG), chromophobic granule (βG) and azurophilic (basophilic) granule (γG) were observed in a single neutrophil. The αG was small (<0.3μm in diameter) and round in shape. The staining pattern of αG was influenced by pH, constituent and concentration of diluent of the staining solution. To estimate the staining condition on αG, five neutrophil types were categorized as follows: type 0, no αG in the cell; type I, αG occupied less than 10% of whole cytoplasm; type II, in 10~40%; type II, in 40~80%; typeIV, more than 80%. Eosinophil of αG was disappeared by longtime Giemsa stain after May-Grunwald. The βG was round to oval and was larger (0.5 μm in diameter) than a G. The γG was round to oval, smaller (<0.4μm in diameter) than βG. To estimate the staining condition on γG, three neutrophil types were classified into as following types as follows: type a, no γG in the cell; type b, γ G occurpied 50% of the whole cytoplasm; type c, in more than 50%. The γG were observed in Giemsa staining preparation, but not in May-Grünwald. However, this granule type appeared by long-time Giemsa stain after May-Grünwald. | |||||||||||
