| 内容記述 | The present study describes the development of an immunodot-blotting technique to determine the sex of Pacific halibut. This method is based on the immunochemical detection of serum vitellogenin existing specifically in females undergoing vitellogenesis. Serum vitellogenin was purified from an estrogen-treated immature females using high-performance liquid chromatography. Two procedures of isolation were combined : gel filtration and anion exchanged chromatography. Isolated vitellogenin has a molecular weight of 380,000 in native form with glycolipophosphoprotein characteristics. Antiserum was raised against estrogen-treated serum, and then, absorbed with male serum to react specifically to estrogen induced proteins including vitellogenin. The developed immunodot-blotting technique by using specific antiserum was effective and rapid determining the sex of Pacific halibut. The evaluated lower limit of this system attained 20ng vitellogenin per dot. | |||||||||||||||||
