| 内容記述 | Mitochondrial (mt-)DNA was isolated from the oocytes of the Japanese oyster, Crassostrea gigas, and a clone for the linearized full-length mt-DNA was obtained by inserting it into the BamHI site of a plasmid vector, pUC118. The total length of the mt-DNA was 17,700 ± 300 base pairs. A primary restriction map has been derived for the 16 cleavage sites made by 8 enzymes, allowing to serve as the basis for the studies of restriction fragment length polymorphism, DNA sequencing and hybridization detection. | |||||

| 内容記述 | マガキ(Crassostrea gigas)の未受精卵からミトコンドリア(mt-)DNAを単離し,制限酵素BamHIで線状化した後,プラスミドベクターpUC 118 に挿入して,mt-DNA全長を含む組換え体のクローンを得た。マガキmt-DNAの長さは17,700±300塩基対であった。8つの制限酵素による16の切断箇所について,概略的な制限酵素切断マップを作成した。得られたマップは,制限酵素切断長多型の研究や DNA 塩基配列の決定,およびハイブリダイゼーションによるmt-DNA断片の検出·同定に有用である。 | |||||
