内容記述タイプ Abstract
内容記述 Since the first occurrence of White Spot Disease (WSD) in 1992, it is still listed as one of the crustacean diseases by the World Organisation for Animal Health in 2022. Horizontal transmission in aquaculture systems is considered as a major contributing factor to the spread of the disease. WSD outbreak was recorded during the experimental run of the co-culture of black tiger prawn (Penaeus monodon) and sandfish (Holothuria scabra) in the grow-out phase. In this study, artificial infection through cohabitation experiments were conducted to determine if H. scabra is a potential vector or reservoir of WSSV. During the first cohabitation (5 days) where WSSV-infected prawn were cohabited with WSSV-free sandfish, 100% of the prawn were one step-positive PCR for WSSV while 100% of the sandfish (coelom, intestine, tentacle, and muscle) were nested-positive PCR for WSSV. Subsequently, WSSV-positive H. scabra from the first cohabitation were transferred to another tank to be cohabited with WSSV-free P. monodon. Sampling of both prawn and sandfish was done every six days post-infection (dpi). At 6 dpi, all prawn and only 1 sandfish were nested-positive for WSSV. At 12 dpi, majority of the prawn were nested-positive and none for sandfish. At 18 dpi, prawn were all negative and one sandfish was nested-positive. At 25 dpi, there were no prawn left due to mortality and 1 of the 3 remaining sandfish was nested positive for WSSV. Based on the results, it was elucidated that sandfish were able to bioaccumulate the viral particles when cohabited with WSSV-infected prawn. Relative to these findings, it was concluded that WSSV is viable in the sandfish due to the consistent nested-positive results of the prawn during the second cohabitation and the occurrence of mortality in the latter part of the experiment. Sandfish could still be safely used for the next co-culture cropping only after quarantine for at least 1 month since the result showed that muscle and intestine can still be WSSV nested-positive after 25 dpi. Hence, sandfish can be a potential reservoir of WSSV for a limited period of time.
